The effect of astragaloside and tanshinone ⅡA on growth factors expression in rat skeletal muscles after acute contusion

Abstract

Objective To observe the effect of astragaloside and tanshinone ⅡA on growth factors expression in rat skeletal muscles after acute contusion. Methods Ninty-six male Wistar rats (weight 180-200 g) were divided into group A (normal control), group B (physiological saline control), group C (astragaloside), group D (tanshinone ⅡA), group E (stragaloside and tanshinone ⅡA mixed injection) and group F (huangqi-danshen injection) randomly (16 rats in each group). In intervention groups, right gastrocnemius of all intervention groups were contused by specific device, then the damaged location was injected with the corresponding drug, and samples were collected from injured muscles after the injury 4, 7, 14 and 28 days. In the normal group, no acute contusion and no drug was injected. Western blot was used to measure the expression of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), hepatocyte growth factor (HGF), insulin-like growth factor-1 (IGF-1) in each group at different time. Results (1) Compared with normal group, the expression of VEGF, bFGF, HGF and IGF-1 in each intervention group increased significantly (P<0.05), which continues until after the injury 28 days. (2) There was no difference of the expression of growth factors among each intervention group (P>0.05). Conclusions Astragaloside and tanshinone ⅡA had no significant effect on growth factors expression in rat skeletal muscles after acute contusion.
